Overview

N-hydroxy-4-(methyl{[5-(2-pyridinyl)-2-thienyl]sulfonyl}amino)benzamide (abbreviated as NHB), is a synthetic small molecule belonging to the sulfanilide and benzamide chemical classes. It has been studied primarily as an inhibitor of histone deacetylase 8 (HDAC8), an enzyme involved in epigenetic regulation through modification of chromatin structure. By inhibiting HDAC8, this compound can modulate gene expression, which may result in anticancer and anti-inflammatory effects, as well as enzyme inhibition activity. The compound has been structurally characterized in complex with human HDAC8 via X-ray crystallography (PDB ID: 1W22)[1][3][5]. There is no evidence that it has reached clinical development or approval for any indication.
